Câu chuyện bắt đầu khi bác sĩ tài năng Matthew Nolan (Richard Armitage) vừa đáp xuống Heathrow sau chuyến bay về từ Bắc Kinh thì lập tức bị bắt vì nghi ngờ liên quan đến cái chết của một phụ nữ Trung Quốc. Dù kêu oan, anh vẫn bị buộc dẫn độ ngay trong đêm để lập tức trở lại Trung Quốc đối mặt với án tử hình. Nhiệm vụ áp giải được giao cho Hana Li (Jing Lusi), nữ thám tử người Anh gốc Á nguyên tắc và cứng rắn, vốn không hề muốn nhận nhiệm vụ này. Cả hai buộc phải lên chuyến bay đêm số hiệu 357 trở lại Trung Quốc.

Nhưng ngay khi máy bay vừa cất cánh, hàng loạt cái chết bí ẩn liên tiếp xảy ra ở độ cao hơn 9.000 mét. Bị mắc kẹt giữa không trung và không còn đường thoát, Nolan và Hana nhận ra họ đang rơi vào một âm mưu chính trị xuyên quốc gia, nơi kẻ sát nhân có thể đang ẩn mình giữa hành khách hoặc phi hành đoàn và sẵn sàng giết người bịt đầu mối.
